It depends. Story-wise it's better to have played the first 3; however, you could probably find a summary or basic overview of the plot to catch you up on that (or if you're an avid reader, they have novels. Assassin's Creed: Renaissance is the book version of Assassin's Creed II). As each Assassin's Creed game introduces something new, there are always tutorials in each game. The only downside is that some of the things from the previous games are more like a review than a tutorial. This game closes the book on Altair and Ezio, two of the characters you play as. I believe it is possible to just pick up Assassin's Creed Revelations and play it, but personally, knowing what I know about the series, I would at least start with Assassin's Creed II.
