It appears to be MDF, but I'm no expert. I can tell you it's not solid wood nor is it cheap particle board.

This is possible, but you would have to drill holes in the appropriate locations to install the rail. It is not pre-drilled on either end.

No. It is preassembled to only be on one side of the bed. Ours was actually on the opposite side than is pictured here.
